Une nuit de 1969, un homme muni d'une importante somme d'argent se rend à l'hôtel El Royale, dont une moitié se situe en Californie et l'autre dans le Nevada. Il y est assassiné par un mystérieux personnage. Dix ans plus tard, le même hôtel accueille différents clients, tous très différents. Il y a notamment le Père Flynn, la chanteuse Darlene Sweet ou encore Laramie Seymour Sullivan, un volubile vendeur d'aspirateurs. Ils croisent bientôt Miles Miller, l'unique employé d'un établissement désormais bien vide.
